The adhesion force of Notch with Delta and the rate of Notch signaling
نویسندگان
چکیده
Notch signaling is repeatedly used during animal development to specify cell fates. Using atomic force microscopy on live cells, chemical inhibitors, and conventional analyses, we show that the rate of Notch signaling is linked to the adhesion force between cells expressing Notch receptors and Delta ligand. Both the Notch extracellular and intracellular domains are required for the high adhesion force with Delta. This high adhesion force is lost within minutes, primarily due to the action of Presenilin on Notch. Reduced turnover or Delta pulling accelerate this loss. These data suggest that strong adhesion between Notch and Delta might serve as a booster for initiating Notch signaling at a high rate.

Comparative evaluation of NOTCH signaling molecules in the endometrium of women with various gynecological diseases during the window of implantation
Objective(s): NOTCH signaling pathway is well known for its role in cell fate, cell survival, cell differentiation, and apoptosis. Some of the NOTCH signaling genes are critical for endometrial function and implantation in animals and appear to play a similar role in humans. The purpose of the current study was to investigate the potential roles of some main components of the NOTCH family in hu...
متن کامل9-cis-Retinoic Acid and 1,25-dihydroxy Vitamin D3 Improve the Differentiation of Neural Stem Cells into Oligodendrocytes through the Inhibition of the Notch and Wnt Signaling Pathways
Background: Differentiating oligodendrocyte precursor cells (OPCs) into oligodendrocytes could be improved by inhibiting signaling pathways such as Wnt and Notch. 9-cis-retinoic acid (9-cis-RA) and 1,25-dihydroxyvitamin D3 (1,25[OH]2D3) can ameliorate oligodendrogenesis. We investigated whether they could increase oligodendrogenesis by inhibiting the Wnt and Notch signaling pathways. Methods: Co...
متن کاملCorrection: An Evolutionary-Conserved Function of Mammalian Notch Family Members as Cell Adhesion Molecules
Notch family members were first identified as cell adhesion molecules by cell aggregation assays in Drosophila studies. However, they are generally recognized as signaling molecules, and it was unclear if their adhesion function was restricted to Drosophila. We previously demonstrated that a mouse Notch ligand, Delta-like 1 (Dll1) functioned as a cell adhesion molecule. We here investigated whe...
متن کاملEndocytic Internalization Routes Required for Delta/Notch Signaling
The internalization of transmembrane receptors from the cell surface plays a central role in signal regulation. Receptor internalization can occur through different routes; however, because of the difficulty in selectively blocking these routes in vivo, their roles in signaling are poorly understood. Here we use null mutations in Drosophila dynamin, clathrin, and AP-2 adaptor subunits to analyz...
متن کاملInfluence of Notch Depth-to-Width Ratios on J-Integral and Critical Failure Load of Single-Edge Notched Tensile Aluminium 8011 Alloy Specimens
In this paper, the influence of notch depth-to-width ratios on J-integral and critical load of Aluminium 8011 alloy specimens with U-notch under Mode I loading are studied. Using experiments, for a set of specimens having different notch depth–to–width ratios, J-integral was found and the same was verified using analytical methods. Further, using the results obtained from the experiments, failu...
